Make sure you use the newly registered users group.
Set that on 1 post. Spammers will (nearly) always give themselves away in the first post.

Check the IP.
If it's from one of the usual suspects like Russia, China, Ukraine, India, Pakistan, Bangladesh, Philippines, Thailand or Vietnam then it's most likely spambots.

setup a good Q&A for registration and that will keep the spam bots out. that 1 post approval settings mentioned above will handle most of the human spammers.

that way you won't have any spammers or their posts to begin with.

to stop the spam bots:

Knowledge Base - How to configure Q&A CAPTCHA.

to stop the human spammers:
admin panel
user registration settings:

set registration to "by user"

then set the "new member post limit" to 1

it is not retroactive, it will only apply to those who register after you set it.

If you want to really get to the nitty gritty check out CIDRAM that I have linked in my signature. Or use the sortables captcha and place all first time posts in moderation queue.
